Dec 21, 2015 at 22:05 comment added NotEvans. sirius-analytical.com/science/pka/methods-measuring-pka ? This kind of method is often used for pharmaceutical compounds, which are weakly acidic/basic to a certain extent. Shows good correspondence with lit. values for pKa of common chemicals too.

Mar 29, 2015 at 22:37 comment added ron There are a number of different spectroscopic techniques that lend themselves to measuring pKa's. One approach involves ion cyclotron resonance. It can accurately assess gas phase equibria. Starting with a molecule with known pKa, add a second molecule with unknown (but expected to be within a few orders of magnitude) pKa and measure the equilibrium constant; work your way up to methane. Other studies have shown a high correlation between gas phase and solution pKa's, see for example pp. 97-98
